Glen Mills has roots dating back to the original land grant given to William Penn in 1681. The area's first settler, George Cheyney, lent his name to the nearby town of Cheyney. Glen Mills got its name from the Willcox family, who constructed two paper mills here—one in 1835 and the other in 1846. From 1864 to 1878, these mills played a crucial role in supplying the United States government with specialized paper for printing government bonds and notes.

While the Glen Mills paper mills have become part of history, the grist mill built by Nathaniel Newlin in 1704 still stands tall, attracting picnickers and history enthusiasts. Additionally, a blacksmith shop, built on the former property in 1975, adds to the area's historical charm. The Newlin Mill Complex's historical significance is acknowledged as it has earned a spot on the National Register of Historic Places.

The arrival of the West Chester and Philadelphia Railroad in Glen Mills around 1858 marked a significant turning point. It provided vital train service connecting Philadelphia and West Chester. Passenger trains operated by SEPTA served Glen Mills until 1986. Today, the West Chester Railroad, a heritage railway, continues to operate trains between Glen Mills and West Chester on weekends.

Over the years, Glen Mills has seen its share of unique attractions, including a Wild West-themed restaurant and roadside attraction known as The Longhorn Ranch during the 1960s. In the 1980s, Pulsations nightclub was a hotspot in the same location. Now, the site is home to a thriving retirement community.

Glen Mills is also known for the bravery of one of its residents, pilot Michael R. Horrocks, who tragically lost his life in the September 11, 2001 attacks. He served as the first officer on Flight 175, the second plane to hit the World Trade Center, and his memory continues to be honored.
